Cleaning and checking carb and petcock will never hurt so I'd recommend you to go ahead. If there's any debris, it should make your bike just run richer afterwards, but at least you know then it's not a petcock.
BTW, you can compare petcock fuel flow between Prime and Run by removing both hoses from the carb. By sucking the vacuum hose you get fuel flow when on Run and you see whether there is any difference compared to Prime. Then you know do you need to touch the petcock at all. I can't even remember if it can be disassembled or not, but you would probably need some new seals which might cost some, so don't remove and open it if you really don't need to.
